Patients with severe renal failure or severe liver dysfunction should avoid using Dulux®.

During the treatment with Darius®, a doctor is recommended to advise patients to limit or avoid excessive exposure to natural or artificial light. Topical calcipotriol can be used in combination with UV radiation only when both the doctor and the patient believe that the potential benefits outweigh the potential risks.
This product contains propylene glycol, which may cause skin irritation.

Calcitriol Ointment Pharmacology and Toxicology

Pharmacological effects:
This product is a topical preparation of vitamin D derivative calcipotriol, which can inhibit the proliferation of skin cells (keratinocytes) and induce their differentiation, thereby correcting the abnormal proliferation and differentiation of psoriatic skin lesions.
Toxicology research:
The effect of this product on calcium metabolism is about 1/100 of the hormone-activated vitamin D3.
Skin cancer studies in mice have shown no particular risk of skin cancer. The calcipotriol solution is 3, 10, and 30 mg / kg / day (in terms of body surface area, 9, 30, and 90 mg / m 2 / day, respectively. It is equivalent to the maximum recommended daily dose of 60 kg for adults 0.25, 0.84 and 2.5 times) topical use for 24 months. High doses are the skin's maximum tolerated dose of calcipotriol. Survival rates were reduced in the 10 and 30 mg / kg / day groups, especially for male rats. Reduced survival is associated with increased incidence of obstructive urinary tract disease. The biggest change related to treatment may be a change in urine composition. This is in line with expectations for the use of high-dose calcipotriol and vitamin D analogs. No effects on the skin, no transdermal or systemic carcinogenesis.
In a study of albino nude mice, repeated exposure to UV radiation and topical use of calcipotriol for 40 weeks, the drug dose level was the same as the previous transdermal carcinogenicity test, and a reduction in skin cancer with reduced UV radiation was observed (Statistically significant in male mice), suggesting that calcipotriol may increase the effect of UV-induced skin cancer. The clinical relevance of these findings is unknown.

Pharmacokinetics of calcipotriol ointment

Kinetic studies show that oral administration is metabolized by the liver and has a short half-life. Human liver homogenate experiments show that human metabolic pathways are similar to those of rats, guinea pigs, and rabbits, and the main metabolites have no pharmacological activity.
Calcipotriol is absorbed through the skin at 1-5% of the administered dose.
