What is Olanzapine?

Olanzapine is a new atypical neuroleptic drug that binds to dopamine receptors, 5-HT receptors and cholinergic receptors and has an antagonistic effect. Antagonism of D2 receptors is associated with positive symptoms of schizophrenia; Antagonism of 5-HT2A receptors is associated with negative symptoms of schizophrenia. Unlike clozapine, this product does not occur with granulocytic deficiency, no delayed onset and severe symptoms of mental depression. Oral absorption is good, food has no effect on its absorption rate, and peak plasma concentration can be reached 5 to 8 hours after oral administration. It is mainly metabolized in the liver, and about 75% of this product is excreted from the urine in the form of metabolites.

Olanzapine indication

It is suitable for the acute phase and maintenance treatment of schizophrenia and other psychosis with severe positive symptoms and / or negative symptoms; it can also alleviate the secondary emotional symptoms common to schizophrenia and related diseases.

Olanzapine clinical application

The oral starting dose, therapeutic dose and maintenance dose are generally 10 mg / day, and the effective range is 5-20 mg / day. The starting dose for elderly patients is 5 mg daily. For patients with severe renal impairment or moderate liver impairment, the starting dose is also 5 mg daily.

Olanzapine adverse reactions

Common adverse reactions are drowsiness and weight gain. Can cause prolactin increase, but it is not related to the dose. Rare side effects include dizziness, headache, dry mouth, constipation, peripheral edema, orthostatic hypotension, delayed extrapyramidal dyskinesia (including Parkinson's syndrome), and transient increase in liver transaminase.

Olanzapine precautions

For severe renal impairment, moderate liver impairment, and elderly patients, the starting dose is halved. Cardiovascular and cerebrovascular patients with a tendency to hypotension, patients with epilepsy, prostatic hypertrophy, patients with paralytic intestinal obstruction, patients with reduced blood cells and neutrophils caused by various reasons, and patients with bone marrow transplantation; use with caution; Use with caution by machine operators.

Olanzapine contraindications
