Progesterone gel pill pharmacology and toxicology
- Progesterone is a steroid hormone secreted by the ovary, placenta and adrenal glands. When sufficient estrogen is present, progesterone can change the endometrium from the proliferative phase to the secretory phase, providing favorable conditions for the implantation of pregnant eggs. After bed, the placenta is formed, which can reduce the excitability of the pregnant uterus and maintain the state of pregnancy; it can promote breast development and prepare for breastfeeding. This product can inhibit the release of luteinizing hormone from the anterior pituitary gland through negative feedback to the hypothalamus, prevent follicles from developing and mature, and inhibit the ovulation process of the ovary.
The mouse's half lethal dose by gavage of this product is 6.2869 (5.2154-7.5785) g / kg, which is equivalent to 388 times the clinical dosage.
Pharmacokinetics of progesterone gel pill
- After oral administration, it is metabolized in the liver, and about 12% is metabolized to pregnanediol. The metabolites are combined with glucuronic acid and excreted in the urine. After oral administration of 100 mg, the blood concentration reached a peak in 2-3 hours, and then gradually decreased, disappearing after about 72 hours, and the half-life was about 2.5 hours.
