When a new gene is identified and its DNA sequence is read, people still can't figure out what the corresponding protein sequence is. This is because without other information, the DNA sequence may be read and translated in six frameworks (three per strand, corresponding to three different start sites). ORF recognition involves detecting these six reading frames and deciding which one contains a DNA sequence bounded by a promoter and a terminator and does not contain a promoter or a terminator within it. A sequence that meets these conditions may correspond to a truly single gene product. Identification of an ORF is a prerequisite to prove that a new DNA sequence is part or all of a specific protein-coding gene.
